Description: CD4 is a 55 kDa type I transmembrane glycoprotein and a member of the Ig-SF of cell surface receptors. It is expressed predominantly on the “helper/inducer” subpopulation of mature T lymphocytes and on monocytes and macrophages. Domains 1 and 2 bind to MHC Class II antigens, while domains 3 and 4 may be involved in cis interactions with the CD3/TCR complex. CD4 functions as an accessory molecule in the recognition of foreign antigens in association with MHC class II antigens by T cells.

Description: CD107b, also known as lysosomal-associated membrane protein 2 (LAMP-2), is a heavily glycosylated type I transmembrane protein that constitutes the major sialoglycoproteins on lysosomal membranes. It is a ligand for galaptin, an S-type lectin present in extracellular matrix, through its recognition of acetyllactosamine oligosaccharide chains and is a ligand for E-selectin-mediated cell adhesion. CD107b is expressed in lysosomal/endosomal membranes and on the surface of activated platelets and some tumor cell lines including U937 and KG1a. LAMP-2 may function in protecting the inner surface of the lysosomal membrane by forming a barrier to lysosomal hydrolases. The upregulation of both CD107a and CD107b on the surface of tumor cell lines has been associated with their enhanced metastatic potential where they may increase adhesion to extracellular matrix and endothelium.

Description: Murine CD38, a type II transmembrane glycoprotein, is a bifunctional ectoenzyme capable of catabolizing nicotinamide adenine dinucleotide (NAD+) to cyclic ADP-ribose (cADPR) and then hydrolyzing cADPR to adenosine diphosphoribose (ADPR). It is expressed at high levels on the surface of peripheral B-lineage cells and at low density on germinal center B cells from unimmunized mice. It has also been reported to be expressed at moderate levels on NK cells, a proportion of peripheral T cells, and a subpopulation of thymocytes which are mostly TCRαβ+, CD4-, CD8-. Murine CD38 is also expressed by all Mac-1+ macrophages in the peritoneal cavities of unimmunized mice, but not by unstimulated bone-marrow-derived macrophages. Description: CD81, also known as TAPA-1, is an integral membrane protein expressed on a variety of cell types and has a high degree of sequence homology between human and mouse. CD81 is expressed on thymic stromal cells where it plays an important role in the transition of γδ+ T cells to more mature T cells with αβ T cell receptors. Immunohistochemical staining has revealed that its expression is localized to the subcapsular region of the thymus and, specifically, on cells that have distinct clustering patterns. It has been speculated that the ligand for CD81 is the pre-T cell receptor which is composed of a TCR β chain and glycoprotein pTα. The monoclonal antibody 2F7 can block thymocyte interaction with CD81 in vitro.
