Description: Polycomb group (PcG) proteins assemble into multimeric protein complexes, which are involved in maintaining the transcriptional repressive state of genes throughout development (1). PHC3 (Polyhomeotic-like protein 3), also known as Early development regulatory protein 3, is a 983 amino acid nuclear protein that is a component of the PcG multiprotein PRC1 complex (2). PRC1 complex acts via chromatin remodeling and modification of histones. PHC3 contains 1 FCS-type zinc finger and SAM (sterile alpha motif) domain (2-3). PHC 3 has been regarded as a candidate tumor suppressor of osteosarcoma (4).

Description: PHOX2A and PHOX2B (Paired mesoderm homeobox protein) are closely related, paired-homeodomain transcription factors that function as determinants of the noradrenergic phenotype during embryogenesis. PHOX2 proteins are crucial for the regulation of endogenous hydroxylases in neural crest cells and promote sympathetic neuron generation. Human PHOX2B contains one DNA binding homeobox domain and is required for the differentiation of all central and nonperipheral noradrenergic centers in the brain. In contrast, PHOX2A controls only the differentiation of the main noradrenergic center of the brain. Regulation of PHOX2 may have therapeutic utility in aging or disorders involving degeneration of noradrenergic neurons.

Description: Signal transducer and activator of transcription 5 (Stat5) belongs to a family of cytoplasmic transcription factors that can be activated (phosphorylated) by a cell surface receptor. Phosphorylation at Tyr694 is obligatory for Stat5 activation. Stat5 has two isoforms, Stat5a and Stat5b. Aberrant Stat5 activation has been implicated in the pathogenesis of chronic myelogenous leukemia, prostate and breast cancer and tumor metastasis. Stat5 is localized in the cytoplasm and upon phosphorylation at Y694 is translocated to the nucleus.

Description: The testis and spermatogenesis cell apoptosis related gene 1 (TSARG1) protein, also known as SPATA3, is highly expressed in testis, and to a lower extent in other tissues. SPATA3 contains a highly conserved J domain, which is present in all DnaJ-like proteins, and is thus a novel member of the HSP40 protein family. SPATA3 is thought to be involved in apoptosis of spermatogenic cells.

Description: IKAP was initially identified as a scaffold protein of the IkB kinase complex that could bind to IKKa, IKKb, NF-kB, and the NF-kB-inducing kinase (NIK), although later evidence has cast doubt on this. More recent reports show that mutations in IKAP such as a frameshift leading to a truncated protein or a missense mutation that leads to defective phosphorylation are responsible for the autosomal recessive genetic disease familial dysautonomia (FD). Reports indicating that it forms part of the RNA polymerase II transcription elongation complex suggest that this disease may be due to compromised transcription elongation. More recently, it was shown that IKAP associates with c-Jun N-terminal kinase (JNK) and could specifically enhance JNK activation induced by the upstream JNK activators MEKK1 and ASK1, indicating another possible cause for FD. At least two isoforms of IKAP are known two exist.

Description: Signal-induced proliferation associated-like protein 2 (SIPA1L3) is a member of the SIPA1 family of RapGAPs. SIPA1L3 was cloned as a novel molecular component of tight junctions and adherens junctions, suggesting that may function to balance Rap1 by counteracting RapGEFs. Recent studies of SIPA indicate that its deregulation can cause myeloproliferative stem cell disorders in mice and increased metastases in human cancers. Other studies suggest SIPA1L1 may play important roles in embryo development and control of cell proliferation. Based on the amount of homology between SIPA family members, it is possible that SIPA1L3 plays other roles development and cell proliferation.

Description: The Silent Information Regulator (SIR2) family of genes are highly conserved from prokaryotes to eukaryotes and have important functions in the regulation of metabolism, growth and differentiation, inflammation, cellular survival, as well as in senescence, lifespan extension and several age-related diseases. Sirtuins are NAD+-dependent histone/protein deacetylases (HDAC) and SIRT3 is the only sirtuin whose increased expression has been shown to correlate with an extended life span in humans. It is localized in the mitochondrial matrix, where it regulates the acetylation levels of metabolic enzymes, including acetyl coenzyme A synthetase 2. SIRT3 is stress-responsive and its increased expression protects myocytes from genotoxic and oxidative stress-mediated cell death.

Description: The interferon-induced protein with tetratricopeptide repeats 3 (IFIT3) protein is a member of a family of tetratricopeptide repeat-containing proteins whose transcription is upregulated by interferons, virus infection, and molecular patterns such as dsRNA or lipopolysaccharides (1,2). These proteins have been suggested to induce anti-viral cellular activities in response to infection (2). IFIT3 has been shown to interact with TBK1 following RNA virus infection, thereby bridging TBK1 to VISA on the mitochondrion and activating IRF3 and thus the immune response (3).
