Description: The monoclonal antibody DX5 reacts with CD49b, also known as very late antigen-2 (VLA-2) and α2 integrin. The antibody stains the majority of NK cells and a small subpopulation of T cells in all mouse strains tested (e.g., A/J, AKR, BALB/c, C3H/HeJ, C57BL/6, C57BL/10, C57BR, C58, CBA/Ca, CBA/J, DBA/1, DBA/2, SJL, SWR). Three-color flow cytometry analysis has revealed that most cells that express NK-1.1 (NKR-P1C) also express CD49b. However, small subsets of DX5+NK-1.1- and DX5-NK-1.1+ cells can be found, especially among the CD3+ population of T lymphocytes. NK cells cultured in the presence of IL-2 progressively lose reactivity with DX5 as a consequence of cellular proliferation. Therefore, DX5 can be used to define functionally distinct subsets of murine NK cells. The monoclonal antibody DX5 has not been demonstrated to have activating or blocking activity.

Description: CD90 (Thy-1) is a GPI-anchored molecule and one of the smallest members of the immunoglobulin superfamily consisting of a single V-set domain. The Thy-1.2 alloantigen is expressed on all thymocytes, peripheral T lymphocytes, and some intraepithelial T cells of most mouse strains. It is also expressed in the brain and at varying levels on other non-lymphoid tissues. Description: CD103 is a member of the integrin series of adhesion molecules. This antigen defines a developmentally important subset of T cells, namely mucosal T cells including all IEL (intraepithelial lymphocytes) and ~20% of lamina propria T cells. Expression of CD103 is more restricted outside these mucosal organs appearing at lower levels on T cell subsets of the lymph node, dendritic epidermis, and periphery. In non-epithelial CD103+ T cells there is a bias toward expression on CD8+ cells. The 2E7 monoclonal antibody exhibits antigen immunoprecipitation patterns similar if not identical to the rat anti-mouse CD103 monoclonal antibody M290. 2E7 is reported to have signal-inducing activity in a redirected lysis assay and to costimulate IEL and CD8+ lymph node cells in conjunction with anti-TCR in vitro which runs counter to the usual inhibition of CTL by anti-integrins. This costimulatory activity is also shared by the monoclonal antibody M290. The property of costimulation is not unique to the mouse since the combination of anti-human αIEL plus anti-TCR will produce similar proliferation in human cells with the exception that CD8+ cells are not stimulated.

Description: CD107a, also known as lysosomal-associated membrane protein 1 (LAMP-1), is a heavily glycosylated type I transmembrane protein that constitutes the major sialoglycoproteins on lysosomal membranes. It is a ligand for galaptin, an S-type lectin present in extracellular matrix, through its recognition of acetyllactosamine oligosaccharide chains, and is a ligand for E-selectin-mediated cell adhesion. CD107a is expressed by activated T cells, macrophages, dendritic cells, activated platelets, tonsillar epithelium, and some tumor cell lines, including U937 and KG1a. It is also a widely expressed intracellular antigen. LAMP-1 may function in protecting the inner surface of the lysosomal membrane by forming a barrier to lysosomal hydrolases. The upregulation of both CD107a and CD107b on the surface of tumor cell lines has been associated with their enhanced metastatic potential where they may increase adhesion to extracellular matrix and endothelium.

Interactions with CD158e1 and certain MHC HLA-B antigens on a target cell inhibits cytotoxicity, possibly preventing target cell lysis and death. KIR and MHC class I interactions may regulate NK and T cell response to antigen stimulation.
