Description: CCR5 antibody detects a G coupled -protein receptor. Human immunodeficiency virus (HIV) and related virus require coreceptors, in addition to CD4, to infect target cells. Some G protein-coupled receptors including CCR5, CXCR4, CCR3, CCR2b and CCR8 in the chemokine receptor family, and four new human molecules GPR15, STRL33, GPR1 and V28 were recently identified as HIV coreceptors. Among them, CCR5 (CC-CKR-5) is a principal coreceptor for macrophage- and dual-tropic HIV-1 strains fusion and entry of human white blood cells. CCR5 is required for the infection by HIV-1, HIV-2, and SIV. The beta-chemokines RANTES, MIP-alpha and MIP-beta are the ligands for CCR5 and prevent infection by M-tropic HIV-1. CXC5 associates with the surface CD4-gp120 of HIV complex and leads to membrane fusion and virus entry of target cells. The amino-terminal domain and the extracellular loops of CCR5 serve as HIV biding sites. CCR5 messenger RNA is expressed in lymphoid organs and monocytes. Description: CD4 is a member of the immunoglobulin superfamily and is implicated as associative recognition element in MHC (Major Histocompatibility Complex) class II-restricted immune response. On T-lymphocytes, it defines the helper/inducer subset. The mature 55 kd CD4 protein consists of a 372 amino acid extracellular segment composed of four tandem immunoglobulin-like VJ regions. The CD4 molecule is a major receptor for human immunodeficiency virus (HIV), binding directly to the envelope glycoprotein gp120 on HIV, with the co-receptors being CCR5 or CXCR4. It has been shown that the V-like domains are critical for binding with HIV envelope gp120.

Description: Apoptosis is related to many diseases and induced by a family of cell death receptors and their ligands. Cell death signals are transduced by death domain containing adapter molecules and members of the caspase family of proteases. These death signals finally cause the degradation of chromosomal DNA by activated DNase. A mouse DNase that causes DNA fragmentation was identified recently and designated CAD (for caspase activated deoxyribonuclease). The human homologue of mouse CAD was more recently identified by two groups independently and termed CPAN and DFF40. Human DFF45 and its mouse homologue ICAD are the inhibitors of CPAN/DFF40 and CAD, respectively. Upon cleavage of DFF45/ICAD by activated caspase, DFF40/CAD is released and activated and eventually causes the degradation of DNA in the nuclei. Activation of CAD/DFF40, which causes DNA degradation, is the hallmark of apoptotic cell death.

Description: BRAL1 is a member a superfamily consisting of several highly homologous hyaluronan and proteoglycan binding link proteins. BRAL1 is predominantly expressed in brain tissue and spinal cord. Like other members in the link-module superfamily, BRAL1 contains an immunoglobulin-like fold and two proteoglycan tandem repeats (PTRs). Its mRNA expression pattern is similar to other lectican proteoglycans, suggesting that BRAL1 may act to stabilize the binding between the extracellular matrix molecule hyaluronan and brevican. Immunostaining of mouse brain showed BRAL1 expression at P20 in the white matter of the developing cerebellum and in myelinated fiber tracts in the adult brain, suggesting that expression starts when axonal myelination occurs.

Description: BRCC45 was initially suggested to be a housekeeping protein that is highly expressed in brain and reproductive organs. Later experiments indicated BRCC45 forms a complex with the breast and ovarian predisposition proteins BRCA1 and BRCA2 as well as RAD51 and BRCC36. This complex has a ubiquitin E3 ligase activity and is thought to enhance cellular survival following DNA damage. BRCC45 has also been suggested to function as a death receptor-associated anti-apoptotic protein by inhibiting the BID-induced activation of the mitochondrial apoptotic pathway. Higher levels of BRCC45 were detected in the majority of hepatocellular carcinomas, suggesting that BRCC45 may promote tumorigenesis when overexpressed.
