Description: Ly-6G/Ly-6C, formerly known as the myeloid differentiation antigen Gr-1, is a GPI-anchored protein. In the bone marrow, the level of antigen expression is directly correlated with granulocyte differentiation and maturation. It is also transiently expressed on monocytes in the bone marrow. In the periphery, Ly-6G/Ly-6C is expressed predominantly on neutrophils.

Description: The immunoglobulin heavy chain binding protein BiP is a member of the hsp70 family of heat shock proteins and is identical to the glucose regulated protein GRP78. While BiP was originally described for its function in B cells, it is now known to be distributed in a variety of tissues, if not ubiquitous. The highly conserved hsp70 proteins have an essential physiological role in stress responses and as “molecular chaperones” which are responsible for a variety of functions such as protein transport, prevention of protein toxicity and direction of protein folding. With regard to its immunological role, BiP is a component of the endoplasmic reticulum and binds free intracellular heavy chains in nonsecreting pre-B cell lines (μ+,L-) or incompletely assembled Ig precursors in H+L+ secreting hybridomas and myelomas. In the absence of light chain synthesis, heavy chains remain associated with BiP and are not secreted. BiP is an ATP binding protein and the dissociation of the BiP-heavy chain complex is probably driven by the ATPase activity attributed to BiP. The monoclonal antibody 76-E6 recognizes a conserved epitope localized within the region of amino acids 497 to 581 of BiP.

Description: The lymphocyte surface antigen CD152, also known as CTLA-4, is related to the costimulatory molecule CD28 and both molecules share common B7 family counter-receptors. However CD152 is thought to be a negative regulator of T cell activation and may play a role in apoptotic control of T cells. CD152 is relatively conserved among humans, mice, and chickens.

Description: CD81, also known as TAPA-1, is an integral membrane protein expressed on a variety of cell types and has a high degree of sequence homology between human and mouse. CD81 is expressed on thymic stromal cells where it plays an important role in the transition of γδ+ T cells to more mature T cells with αβ T cell receptors. Immunohistochemical staining has revealed that its expression is localized to the subcapsular region of the thymus and, specifically, on cells that have distinct clustering patterns. It has been speculated that the ligand for CD81 is the pre-T cell receptor which is composed of a TCR β chain and glycoprotein pTα. The monoclonal antibody 2F7 can block thymocyte interaction with CD81 in vitro.

Description: The CD45 antigen is an essential regulator of leukocyte activation and development and is now known to be a protein tyrosine phosphatase, e.g., CD45 is able to dephosporylate the Src family of protein kinases. This activity is known to be required for signal transduction induced by T and B cell antigen receptor engagement, cytokine signaling, and Fc receptor stimulation. The functional state of CD45 also has a pronounced effect on lymphocyte development. CD45 exists in multiple forms, such as the restricted variants (CD45R; see companion products 1660, 1665, 1675, 1685 and 1820) and as allelic forms CD45.1 (Ly-5.1) and CD45.2 (Ly-5.2). The specificity of antibodies to CD45.1 and CD45.2 has been exploited in studies where resolution of donor and recipient cells is essential, such as in gene therapy or bone marrow transplantation. The monoclonal antibody 104 recognizes CD45 (leukocyte common antigen) on all leukocytes of mouse strains expressing the CD45.2 allotype (e.g., A, AKR, BALB/c, CBA/Ca, CBA/J, C3H/He, C57BL, C57BR, C57L, C58, DBA/1, DBA/2, NZB, SWR, 129).

Description: CD11a, also known as leukocyte function-associated antigen-1 (LFA-1α), represents the 180 kDa integrin αL subunit which combines with the CD18 β2 integrin subunit to form the αLβ2 integrin heterodimer. It is expressed on lymphocytes, granulocytes, monocytes, and macrophages and is upregulated on activated T cells. CD11a mediates adhesion of lymphocytes to vascular endothelium and is involved in costimulation.

The former arises as a result of alternative splicing of different exons. The specificity of antibodies to CD45.1 and CD45.2 has been exploited in studies where resolution of donor and recipient cells is essential, such as in gene therapy or bone marrow transplantation. The monoclonal antibody A20 recognizes all mouse leukocytes expressing the CD45.1 allotype.

Description: Murine CD38, a type II transmembrane glycoprotein, is a bifunctional ectoenzyme capable of catabolizing nicotinamide adenine dinucleotide (NAD+) to cyclic ADP-ribose (cADPR) and then hydrolyzing cADPR to adenosine diphosphoribose (ADPR). It is expressed at high levels on the surface of peripheral B-lineage cells and at low density on germinal center B cells from unimmunized mice. It has also been reported to be expressed at moderate levels on NK cells, a proportion of peripheral T cells, and a subpopulation of thymocytes which are mostly TCRαβ+, CD4-, CD8-. Murine CD38 is also expressed by all Mac-1+ macrophages in the peritoneal cavities of unimmunized mice, but not by unstimulated bone-marrow-derived macrophages. Monoclonal antibodies to CD38 have been shown to induce B and T cell proliferation, protect B cells from apoptosis, and inhibit B lymphopoiesis.
