Description: Proteins that directly bind to nascent RNA polymerase II transcripts, the heterogenous nuclear ribonucleoproteins (hnRNPs), play an important role in both transcript-specific packaging and alternative splicing of pre-mRNAs. A group of abundant hnRNPs, the M1-M4 proteins, appear as a cluster of four proteins. The M proteins are pre-mRNA binding proteins in vivo, and they bind avidly to poly (G) and poly (U) RNA homopolymers in vitro. M proteins are members of the ribonucleoprotein consensus sequence family of RNA-binding proteins with greatest similarity to a hypothetical RNA-binding protein from Saccharomyces cerevisiae. The M proteins also possess an unusual hexapeptide-repeat region rich in methionine and arginine residues (MR repeat motif) that resembles a repeat in the 64kDa subunit of cleavage stimulation factor, which is involved in 3'-end maturation of pre-mRNAs. Proteins immunologically related to M exist in divergent eukaryotes ranging from human to yeast.

Description: Eukaryotic translation initiation factor 2C (eIF2C) proteins (argonaute family) influence RNA interference (RNAi) as components of the RNA-inducible silencing complex (RISC) or microRNA (miRNA)-containing ribonucleoprotein particle (miRNP). Small RNAs, including small interfering RNAs (siRNAs) and miRNAs, can silence target genes through mechanisms that utilize RISC or miRNP particles. eIF2C1 (argonaute 1, AGO1, eIF2C, GERP95, Q99) and Dicer1 play a coordinated role in siRNA-mediated gene silencing. eIF2C2 (Slicer, argonaute 2, AGO2, Q10) is a RISC component that can concentrate in cytoplasmic processing bodies (P-bodies) and catalyze mRNA cleavage. Mammalian P-bodies contain mRNAs and have an association with miRNA-induced translational silencing and siRNA-induced mRNA degradation. Additional eIF2C proteins include eIF2C3 (argonaute 3, AGO3), eIF2C4 (argonaute 4, AGO4) and meIF2c5 (mouse argonaute 5).

Description: SIP1 Antibody: SIP1 is one of the proteins found in the SMN complex, which consists of the survival of motor neuron (SMN) protein and several gemin proteins. The SMN complex is localized to a subnuclear compartment called gems (gemini of coiled bodies) and is required for assembly of spliceosomal snRNPs and for pre-mRNA splicing. SIP1 interacts directly with the SMN and it is required for formation of the SMN complex. A knockout mouse targeting the mouse homolog of this gene exhibited disrupted snRNP assembly and motor neuron degeneration. However, knockdown of the SIP1 mRNA in motor neurons showed normal motor axons while that of SMN mRNA did show abnormal motor axon outgrowth, indicating that SIP1 may have additional roles outside of the SMN complex.

Description: hnRNP F is part of the subfamily of ubiquitously expressed heterogeneous nuclear ribonucleoproteins (hnRNPs). These are RNA binding proteins that complex with heterogeneous nuclear RNA (hnRNA) and appear to influence pre-mRNA processing and other aspects of mRNA metabolism and transport. hnRNP F binds to RNAs which have guanosine-rich sequences.

Description: Eukaryotic translation initiation factor 4E is a 217 amino acids protein that belongs to the eukaryotic initiation factor 4E family. eIF4F is a multi-subunit complex, the composition of which varies with external and internal environmental conditions. It is composed of at least EIF4A, EIF4E and EIF4G1/EIF4G3. EIF4E is also known to interact with other partners. It can recognize and bind the 7-methylguanosine-containing mRNA cap during an early step in the initiation of protein synthesis and facilitates ribosome binding by inducing the unwinding of the mRNAs secondary structures.

Description: Heterogeneous nuclear ribonucleoproteins (hnRNPs) are thought to be involved in pre-mRNA processing. However, its role in the regulation of gene expression is as yet poorly understood. Proteins of the heterogeneous nuclear ribonucleoparticles (hnRNP) family form a structurally diverse group of RNA binding proteins implicated in various functions. Recently, hnRNP proteins have been shown to hinder communication between factors bound to different splice sites. Conversely, several reports have described a positive role for some hnRNP proteins in pre-mRNA splicing. hnRNP-U, also termed scaffold attachment factor A (SAF-A), binds to pre-mRNA and nuclear matrix/scaffold attachment region DNA elements.

Description: DEAD-box proteins, characterized by the conserved motif Asp-Glu-Ala-Asp, are putative RNA helicases implicated in several cellular processes involving modifications of RNA secondary structure and ribosome/spliceosome assembly. Based on their distribution patterns, some members of this family may be involved in embryogenesis, spermatogenesis, and cellular growth and division. DDX15 (DEAH-box protein 15), also known as DHX15, DBP1 or HRH2, is a nuclear ATP-dependent RNA helicase that is involved in pre-mRNA splicing and is a member of the DEAH-box subfamily of DEAD-box proteins. Expressed throughout the body, DDX15 is a pre-mRNA processing factor that plays a role in spliceosome disassembly after the release of mature mRNA. When localized to the nucleoli, DDX15 is thought to interact with the La/SSB autoantigen, an RNA chaperone that functions in various intracellular processes. DDX15 is 795 amino acids in length and is the human ortholog of the S. cerevisiae protein Prp43. helicase 2.

Description: The exosome is a multisubunit complex composed of several highly conserved subunits, some of which are 3? to 5? exoribonucleases. The complex is involved in a variety of cellular processes and is responsible for degrading unstable mRNAs that contain AU-rich (ARE) elements in their untranslated 3? region. DIS3L2 (DIS3-like exonuclease 2) is an 885 amino acid protein that is thought to function as an exonuclease and may be required for the 3? processing of pre-mRNA into mature mRNA. Defects or chromosomal translocations involving the gene encoding DIS3L2 may be associated with Marfanoid habitus, a genetic disorder characterized by abnormalities in the skeleton, eyes and cardiovascular system. DIS3L2 is expressed as five isoforms due to alternative splicing events.
