Description: STING A162 variant (human recombinant) contains amino acids 138-379 of the wild-type variant (R232) with an alanine substituted for serine at position 162. Stimulator of interferon genes (STING) is a component of the innate immune response that binds to cyclic dinucleotides, which are bacterial second messengers, leading to activation of NF-κB and transcription of immunomodulatory genes, including type I interferon (IFN). The R232 variant of STING is the most common variant in the human population, found at a frequency of 57.9% in the 1000 Genome Project. The SNP variant H232 is found at a 13.7% frequency. The S162A point mutation is located in the cyclic dinucleotide binding site of the human STING variants R232 and H232 and allows human STING to bind to DMXAA, a compound previously known to bind mouse, but not human, STING. When STING S162A is bound to DMXAA, it adopts the closed conformation, similar to the conformation it has when bound to the second messenger 2’3’-cGAMP (Item No. 19887), and activates the IFN pathway similarly to mouse STING.

Description: Cyclic GMP-AMP (cGAMP) synthase (cGAS; Item No. 22810) is a nucleotidyltransferase located in the cytosol that acts as a cytosolic DNA sensor to detect foreign DNA from microbial pathogens as part of the innate immune response. Upon binding to cytosolic DNA, cGAS produces the cyclic dinucleotide second messenger cGAMP, which activates stimulator of interferon genes (STING), leading to activation of the type I interferon (IFN) pathway. <em>In vitro</em>, fibroblasts, macrophages, and dendritic cells isolated from cGAS knockout (cGAS<sup>-/-</sup>) mice do not produce type I IFNs following DNA transfection or DNA virus infection.{39163} Similarly, cells containing a frame-shift mutation in the cGAS locus fail to mount an immune response to HIV and other retroviruses.{39164} <em>In vivo</em>, cGAS<sup>-/-</sup> mice infected with herpes simplex virus 1 (HSV-1) have lower levels of IFN-α and IFN-β, shorter survival times, and higher post-mortem levels of HSV-1 in the brain. TBK1 also phosphorylates the autophagy receptor optineurin to enhance binding of ubiquitin-like microtubule-associated protein light chain 3 (LC3) modifiers and induce autophagic clearance of <em>S. enterica</em>.

ML RR-S2 CDA increases type I interferon production by THP-1 human monocytes relative to unmodified cyclic di-AMP (CDA; Item No. 17753), indicating the ML enhances its action at human STING. It induces expression of IFN-β and the pro-inflammatory cytokines TNF-α, IL-6, and Mcp-1 in murine bone marrow macrophages (BMM) isolated from wild-type, but not STING-/-, mice. ML RR-S2 CDA also induces IFN-β expression in peripheral blood mononuclear cells (PBMCs) isolated from donors carrying STINGWT/WT, STINGWT/REF, and STINGWT/HAQ alleles. In vivo, ML RR-S2 CDA initiates tumor regression and prevents tumor growth upon tumor cell reimplantation in 4T1 breast and CT26 colon cancer mouse xenograft models.
