Description: CD5, also known as Lyt-1, is a monomeric type I transmembrane glycoprotein expressed on thymocytes, T lymphocytes, and a subset of B lymphocytes, but not on natural killer (NK) cells. It has been identified as the major ligand of the B-cell antigen CD72. The frequency of CD5+ B cells exhibits strain-dependent variation, and the phenotypic, anatomical, functional, developmental, and pathological characteristics of the CD5+ B cells suggest that they may represent a distinct lineage, known as B-1 cells. Binding of CD5 on the T cell surface can augment alloantigen- or mitogen-induced lymphocyte proliferation and induces increased cytosolic free calcium, IL-2 secretion, and IL-2R expression. It has been proposed that CD5 negatively regulates signal transduction mediated by the T-cell and B-cell receptors.

The 145-2C11 monoclonal antibody recognizes an epitope on the 25 kDa ε chain of the CD3/TCR complex. In the presence of Fc receptor-bearing accessory cells, soluble 145-2C11 can activate primed and naïve T cell in vitro. 145-2C11 can also induce redirected lysis of specific target cells by CTL clones and it can block lysis of specific target cells by antigen-specific CTL’s. Immobilized 145-2C11 can activate both normal T lymphocytes and cloned T cell lines. Under certain conditions, T cell activation by 145-2C11 may result in apoptotic cell death.

Description: Chicken CD45 is a transmembrane glycoprotein expressed on all leukocytes. B lineage cells in the bursa of Fabricius express the 215 kDa isoform while T lineage cells bear the 190 kDa variant. These high molecular weight molecules have intrinsic phosphotyrosine phosphatase activity characteristic of mammalian CD45. Levels of CD45 expression detected by the monoclonal antibody LT40 in the avian thymus are heterogeneous with approximately 90% of thymocytes expressing fourfold higher levels of surface CD45 (CD45hi) than do the remaining 10% (CD45low) of thymocytes. The CD45low population contains exclusively thymocytes with the CD3-CD4-CD8low phenotype characteristic of the immediate precursors to the CD3-CD4+CD8+ thymic population which are CD45hi. This shift from low to high levels of surface CD45 expression is concomitant with the transition from CD4-CD8low to CD4+CD8+ and before the expression of CD3. The ability of the monoclonal antibody LT40 to discriminate this CD45low subpopulation provides a suitable marker for the CD3-CD4-CD8low immediate precursors to the CD3-CD4+CD8+ thymocytes.
