Description: The hybridoma secreting the antibody to subunit β2 was generated by fusion of splenocytes from Balb/c mice which had received repeated immunization with dinitrophenylated proteasomes. The antibody (clone MCP165) has been characterised by one- and two dimensional Western blotting. The antibody has not been demonstrated to immunoprecipitate the native 20S proteasome. The immunoglobulin subclass is IgG1.

Supplier: Tonbo Biosciences
Description: The 53-6.7 antibody reacts with the 32-34 kDa alpha subunit of mouse CD8, known as CD8a or CD8 alpha. CD8a can form a homodimer (CD8 alpha-alpha), but is more commonly expressed as a heterodimer with a second chain known as CD8b or CD8 beta. CD8 acts as a co-receptor in antigen recognition and subsequent T cell activation that is initiated upon binding of the T cell receptor (TCR) to antigen-bearing MHC Class I molecules. The cytoplasmic domains of CD8 provide binding sites for the tyrosine kinase lck, facilitating intracellular signaling events that lead to T cell activation, development, and cytotoxic effector functions. CD8+ cytotoxic T cells (CTLs) play an important role in inducing cell death of tumor cells, as well as cells infected by virus, bacteria or parasites.

Supplier: Tonbo Biosciences
Description: The 3G3 antibody reacts with mouse Foxp3, a 50-55 kDa transcription factor which is a central regulator of T cell activity and is critical for the development and function of regulatory T cells (Tregs). Foxp3 is expressed at constitutively high levels in Treg cells, which are further identified as being CD4+ CD25+. In resting conventional T cells (CD4+ CD25-) Foxp3 expression is restricted, and upon TCR activation is expressed only transiently and in a small proportion of cells. However, the growth factor TGF-beta has been shown to induce expression of Foxp3 in naïve T cells, driving their development into Foxp3+ Tregs, which are called “induced” or “adaptive” Tregs. These cells are phenotypically similar to so-called “natural” Tregs (CD4+ CD25high Foxp3+) which originate in the thymus and comprise the majority of Treg cells. Tregs are critical for maintaining peripheral tolerance and are implicated in the development of autoimmunity.

Description: The OKT4 antibody reacts with human CD4, a 59 kDa protein which acts as a co-receptor for the T cell receptor (TCR) in its interaction with MHC Class II molecules on antigen-presenting cells. The extracellular domain of CD4 binds to the beta-2 domain of MHC Class II, while its cytoplasmic tail provides a binding site for the tyrosine kinase lck, facilitating the signaling cascade that initiates T cell activation. CD4, and co-receptors CCR5 and CXCR4, may also be utilized by HIV-1 to enter T cells. Human CD4 is typically expressed on thymocytes, some mature T cell populations such as Th17 and T regulatory (Treg) cells, as well as on dendritic cells.

Description: The 15.2 antibody reacts with human CD54, also known as ICAM-1 (Intercellular Adhesion Molecule 1), a 90-110 kDa cell surface glycoprotein that is inducibly expressed on both immune and endothelial cells. As its name implies, ICAM-1 participates in cell-cell adhesion between leukocytes and endothelial cells, facilitating leukocyte recruitment and transmigration at sites of inflammation. The ligands for ICAM-1 are also expressed on leukocyte and endothelial cells, and include Mac-1, fibrinogen, and a member of the integrin protein family, LFA-1 (CD11a).

Description: ImmunoReagents’ Alkaline Phosphatase (AP) antibody conjugates are used in ELISA, Immunohistochemistry (IHC) and Western blotting techniques. AP is a 140 kDa enzyme that changes color in the presence of PNPP or BCIP/NBT substrates. These conjugates have a linear reaction rate thus allowing the researcher to increase sensitivity by prolonging substrate incubation times.

Description: The 70 kDa heat shock protein Hsp70 belongs to the Hsp70 family of highly-related protein isoforms ranging in size from 66 kDa to 78 kDa. Hsc70 shares close biochemical and biological ties to Hsp70, and also belongs to the Hsp70 family. These proteins include cognate members found within major intracellular compartments and highly inducible isoforms predominantly cytoplasmic or nuclear in distribution. Members of the Hsp70 family function as molecular chaperones involved in such cellular functions as protein folding, transport, maturation and degradation, operating in an ATP-dependent manner. The molecular chaperones of the Hsp70 family recognize and bind to nascent polypeptide chains or partially folded intermediates of proteins, preventing their aggregation and misfolding, and the binding of ATP triggers a critical conformational change leading to the release of the bound substrate protein. Data demonstrates that with a ubiquitin-like domain at its amino terminus and its association with the 26S proteosome in HeLa cells, Bag-1 modulates the chaperone activity of Hsc70 and Hsp70. These findings reveal Bag-1's role as a physical link between the Hsc70/Hsp70 chaperone system and the proteasome. Experimental data also shows that the ATPase domain and the substrate-binding domain of Hsp70 (or Hsc70) cooperate to form a co-chaperone-chaperone complex with the synaptic vesicle cysteine string protein (csp), essential for normal neurotransmitter release.

Supplier: Tonbo Biosciences
Description: The IM7 antibody recognizes CD44, a ubiquitously expressed cell surface receptor which is important for extracellular matrix organization, cell-cell and cell-matrix adhesion and migration. CD44 may be expressed in a number of different isoforms (splice variants) from the most typical or “standard” form, known as CD44s, to variants designated CD44v, e.g. CD44v1 or CD44v6. These receptors interact with several ligands, but most often associate with an extracellular matrix component hyaluronate, through which it mediates adhesion.

Description: Ghrelin is a multifunctional peptide hormone secreted from the anterior pituitary known to affect many physiological processes including the release of growth hormone, appetite regulation, gastric acid secretion, gastrointestinal motility, and blood pressure regulation. The 28 amino acid ghrelin peptide is modified at Ser3 by addition of a fatty acid group (i.e., n-octanoyl) that is essential for its biological activity. The ghrelin receptor (GHS-R) is a seven transmembrane (7-TM) GPCR that is expressed in the hippocampus, pituitary, and hypothalamus, as well as in peripheral tissues including the heart, lung, liver, kidney, pancreas, intestines, and immune cells. Ghrelin signals are primarily relayed via coupling of the GHS-R to Gq/11 alpha subunits, promoting stimulation of phospholipase C, and an increase in intracellular calcium. Stimulation of adenylyl cyclases and increased cAMP are also induced by GHS-R activation. In humans and swine, two splice variants of the ghrelin receptor are expressed from the same gene. The GHS-R 1(a) receptor is the 366 amino acid, 7-TM form, while GHS-R 1(b) is a 289 amino acid, 5-TM form that is truncated at the C-terminus and is pharmacologically inactive. The epitope recognized by this antibody is unaffected by alternative splicing.
