G-Biosciences' IBS™ Buffer kit is specifically developed for gentle solubilization of inclusion bodies and contains a simple protocol where inclusion bodies are suspended in IBS™ Buffer and readily dissolved, releasing the proteins of interest. Once the inclusion bodies are solubilized, the sample is ready for further analysis and other downstream applications.
The chemical composition of the supplied IBS™ Buffer and denaturant are compatible with widely used protein folding protocols. IBS™ Buffer is also compatible for protein estimation with G-Biosciences' NI™ Protein Assay (Supplier No. 786-005) and for running SDS-PAGE analysis using the PAGE-Perfect™ kit (Supplier No. 786-123).
For solubilization of hydrophobic proteins from inclusion bodies, G-Biosciences offers the IBS-HP™ Buffer, which contains optimized concentrations of SDS, a buffering agent and urea. After solubilization of inclusion bodies, free SDS may be quenched or removed by competing with a non-ionic detergent. The kit is supplied with optional DTT.
A major problem with bacterial expression is that non-bacterial and over-expressed proteins form aggregates of insoluble proteins, known as inclusion bodies, especially when expressed at high levels. The aggregated proteins are difficult to solubilize, due to the nature of aggregates; however, G-Biosciences offers a selection of products for dealing with the range of issues involved with solubilizing and recovering active proteins from inclusion bodies.
