Peripheral blood cells are collected from healthy donors by direct collection or by using the Spectra Optia® Apheresis system. Cell-rich leukapheresis product is extracted using continuous flow centrifugal technology directly into the collection bag.
Purification, isolation, and cryopreservation of cells are performed on-site immediately following collection utilizing best-in-class instruments, biopreservation media, and standardized protocols. The rigorous operational controls mitigate any potential loss while ensuring viable and functional cells post-thaw, guaranteeing that customers receive consistent high-quality cells.
Mobilized leukopaks: Mobilized leukopaks are derived from mobilized peripheral blood and are an excellent source of CD34+ hematopoietic stem and progenitor cells (HSPCs), which are used as starting material for transplantation, regenerative medicine, and cell-based therapies.
Dosing regimens for mobilized leukopaks: Mobilized leukopaks are collected using the Spectra Optia® system into a collection bag containing ACD-A anticoagulant. G-CSF, Plerixafor, and Dual (G-CSF+Plerixafor) mobilization regimens are available for leukopak collections. Along with choosing the mobilization agent(s) that best suits the downstream application, researchers can also select different dosing regimens that include varying mobilization durations and collection days as detailed in the dosing regimen table. Mobilization agents, dosage, dosing schedule, and number of collections all impact total stem cell yields. In general, increasing the mobilization duration and including dual mobilization regimes (G-CSF+Plerixafor), in conjunction with multiple day collections, will result in substantial benefits in terms of CD34+ stem cell yields in the mobilized leukopaks.
In practice, adherence to mobilization regimens is always dependent on donor tolerance to the specific regimen requested. Due to the potential side effects of these agents and the complexity surrounding donor recruitment and collection, donors must be monitored closely, and collection must be managed continuously to ensure both donor safety and optimal cell collection from the mobilized peripheral blood.
