Authenticase™ is a proprietary mixture of structure-specific nucleases capable of recognizing and cleaving outside mismatch and indel (insertion and/or deletion) regions, ranging from 1 to 10 basepairs (bp) on double-stranded DNA.

Authenticase is a mixture of enzymes capable of recognizing mismatch and indel regions on a double-stranded DNA and creating double-stranded cleavages around these regions and with a limited non specific activity on homoduplex regions of DNA. The regions of mismatch or indel could range from at least 1 to 10 bps. The activity of cleavage on mismatch/Indel regions can be utilized in the process of oligo-based PCR gene synthesis to improve the accuracy of PCR amplicons. Alternatively, Authenticase™ treated products can be analyzed to determine the popularity of heteroduplex of a PCR pool amplified from enzyme-based edited genome.
