In these activities, students play the role of the diagnostician in the following scenario. Patients arrive at the emergency room with fever and cough. A viral infection is suspected and must be confirmed or ruled-out from patient samples. Pre-amplified plasmid DNA is used to emulate the virus; no actual viral particles are used.
In both kit versions, students will perform a simulated loop-mediated isothermal amplification (LAMP) reaction and will verify DNA amplification occurred by observing a color change. Both kits contain enough reagents for 24 reactions and must be stored in the freezer upon arrival). Simulated LAMP kit is also available elsewhere.
Iso-B kit LAMP Reaction and verification can be done without any specialized equipment other than a micropipette or capillary pipette.
Iso-S Kit LAMP reaction and verification can be done without any specialized equipment other than a micropipette or capillary pipette. This kit also incorporates a gel electrophoresis step which allows students to visualize the DNA after performing the LAMP reaction. For this step, standard DNA Gel electrophoresis equipment will be required.
